چکیده

An increase in cardiac P-adrenergic sensitivity or 8-receptor density or both has been described in several animal species after denervating the heart. The transplanted human heart is also denervated and, therefore, may exhibit supersensitivity to /3-adrenergic agonists and an increase in .3-adrenergic receptor density. In 16 patients examined 1-3 months after orthotopic cardiac transplantation, /3-adrenergic receptor density measured by [t211]iodocyanopindolol binding in endomyocardial biopsy specimens was not significantly different in transplant recipients compared with normal controls (transplant=1,429±199, control=1,728±263 fmol/g wet wt; p=NS). However, when normalized to Lowry protein, the ['25lliodocyanopindolol in ,8-adrenergic receptor density in biopsy tissue from transplant recipients was significantly lower than in tissue from controls (transplant=58.1+6.2, control=93.5+13.4 fmol/g Lowry protein;

Isolated presynaptic inotropic beta-adrenergic supersensitivity of the transplanted denervated human heart in vivo.

BACKGROUND The regulation of contractility of the transplanted heart depends on circulating catecholamines resulting from cardiac denervation. Supersensitivity to circulating catecholamines may result from loss of presynaptic neuronal uptake or upregulation of postsynaptic beta-adrenergic receptors. METHODS AND RESULTS Dose-response curves using the beta-adrenergic receptor agonists isoprenal...

f-Adrenergic Receptor Number and Adenylate Cyclase Function in Denervated Transplanted and Cardiomyopathic Human Hearts

To test the hypothesis that there is up-regulation of /-adrenergic receptor density or supersensitivity of ,-adrenergic receptor-stimulated adenylate cyclase in the denervated transplanted human heart, we studied myocardium from transplanted, normal, and failing hearts. Myocardium was obtained from 10 patients 9±3 months after cardiac transplantation, from 10 patients without cardiac disease, a...

Mechanisms of supersensitivity to sympathomimetic amines in the chronically denervated heart of the conscious dog.

Mechanisms of denervation supersensitivity to sympathomimetic amines were studied in conscious animals. Norepinephrine, 0.1 micrograms/kg, increased left ventricular dP/dt significantly more (4208 +/- 828 mm Hg/sec) in dogs with cardiac denervation than in intact dogs (1029 +/- 280 mm Hg/sec), P less than 0.01, whereas responses to isoproterenol were similar in both groups. Denervation supersen...
